Description

Grapevines are easy to be attacked by Erysiphe necator, so it is significant to uncover why Chinese wild Vitis pseudoreticulata has strong resistance to such a common fungi, which can provide guidance for breeding disease resistance grapevine varieties. This project is aimed to unravel the defense mechanism of Chinese wild Vitis pseudoreticulata to Erysiphe necator on posttranscriptional level mediated by microRNAs. To profile the microRNA expression affected by Erysiphe necator infection and identify microRNAs relative to powdery mildew resistance, two sRNA libraries were constructed from inoculated and non-inoculated grapevine leaves for small RNA high throughput sequencing.

Neuro-2a cells were infected with ZIKV (MOI = 0.5) for 48 h and total RNA was extracted and purified using TRI Reagent and RNeasy Mini kit (Qiagen). RNA-seq was performed at the Molecular and Genomics Core Facility of the University of Mississippi Medical Center. Differential expression analysis between mock and ZIKV infected cells was done using Tophat and cuffdiff programs from the tuxedo suite.
